杏仁甙主要存在于苦杏、苦扁桃、桃、油桃(nectarines)、枇杷、李子、苹果、黑樱桃等果仁和叶子中，苦杏仁皮中不含苦杏仁甙。

苦杏仁甙由龙胆二糖和苦杏仁睛组成的β-型糖苷，分子式：C20H27NO11，分子量：457.43。

其三水合物为斜方柱状结晶，熔点200，无水物熔点约有20，[α]：20D-42。1g可溶于12mlH2O，900ml乙醇及11ml乙醇，易溶于沸水。

苦杏仁中苦杏仁甙的含量约3%，是甜杏仁高20~30倍，其中，皮尖部含量略高，皮中不含苦杏仁甙。生杏仁含HCN0.179%，煎煮损耗98%的氢氰酸（HCN，H-C≡N），炒制损耗3-7%的HCN。

苦杏仁中蛋白质包括分解苦杏仁甙的酶，需要在制备过程中使其失去活性，常用焯法，方法是水沸（90-100）后，将苦杏仁（带皮）放入煎煮，宜掌握时间长短要适宜（一般约5分钟，太长则丢失原量的34.1%左右，太短则不足以使苦杏仁甙酶失去活性。），取出后，脱皮，烘干，再次煎煮。

苦杏仁中含脂肪酸50%，为高级润滑油，可食用，主要是亚油酸（占27%）、油酸（67%）、棕榈酸（5.2%），具有驱虫、杀菌、润肠和软化皮肤角质层等作用，但制备苦杏仁甙时需要除去，出油率为44%。
